A general nonparametric framework for testing hypotheses about function-valued parameters

We present a general nonparametric approach for testing whether a statistical parameter defined through conditional distributions is constant across the conditioning variables. Such hypotheses arise naturally in problems such as assessing treatment effect heterogeneity, conditional associational effects, and conditional mean dependence. Our framework studies function-valued parameters obtained by evaluating a smooth statistical functional on conditional probability distributions. We establish an explicit connection between our test and procedures based on studying the norm of the function-valued parameter. Unlike many existing norm-based tests, which exhibit poor asymptotic behavior under the null, the proposed test statistic admits a tractable limiting null distribution. We illustrate the applicability of the proposed test through several examples, assess its operating characteristics in simulation studies, and apply it to data from a breast cancer trial to identify predictive biomarkers for response to adjuvant chemotherapy.

Generalized projection tests for function-valued parameters with applications to testing structural causal assumptions

Structural assumptions are central to the causal inference literature. In practice, it is often crucial to assess their validity or to test implications that follow from them. In many settings, such tests can be framed as evaluating whether a function-valued parameter equals zero. In this paper, we propose a class of generalized projection tests based on series estimators for function-valued parameters. We establish conditions under which the proposed tests are valid and illustrate their applicability through examples from the data fusion and instrumental variables literature. Our approach accommodates flexible machine learning methods for estimating nuisance parameters. In contrast to many existing approaches, the limiting distribution of the proposed test statistics is straightforward to compute under the null hypothesis. We apply our method to test the equality of conditional COVID-19 risk across vaccine arms in the COVID-19 Variant Immunologic Landscape (COVAIL) trial.

Developing Performance-Guaranteed Biomarker Combination Rules with Integrated External Information under Practical Constraint

In clinical practice, there is significant interest in integrating novel biomarkers with existing clinical data to construct interpretable and robust decision rules. Motivated by the need to improve decision-making for early disease detection, we propose a framework for developing an optimal biomarker-based clinical decision rule that is both clinically meaningful and practically feasible. Specifically, our procedure constructs a linear decision rule designed to achieve optimal performance among class of linear rules by maximizing the true positive rate while adhering to a pre-specified positive predictive value constraint. Additionally, our method can adaptively incorporate individual risk information from external source to enhance performance when such information is beneficial. We establish the asymptotic properties of our proposed estimator and compare to the standard approach used in practice through extensive simulation studies. Results indicate that our approach offers strong finite-sample performance. We also apply the proposed methods to develop biomarker-based screening rules for pancreatic ductal adenocarcinoma (PDAC) among new-onset diabetes (NOD) patients.
