A Zero-Inflated Beta Mixture Model for Marginal Mediation Analysis with Compositional Microbiome Mediators

The role of the microbiome in disease pathogenesis is an emerging field with strong evidence suggesting that dysbiosis is associated with precancerous and cancerous states. Microbiome data present substantial challenges for causal mediation analysis due to sparsity, compositional constraints, and latent heterogeneity. To address these issues, we propose a zero-inflated beta mixture (ZIBM) method for mediation analysis with compositional microbiome mediators. The proposed method accommodates excess zeros through a zero-inflation component and captures heterogeneity in non-zero relative abundances using a beta mixture distribution. Within the potential-outcomes framework, the ZIBM provides estimates of marginal microbiome-mediated causal effects, and model parameters are estimated using an expectation-maximization algorithm. Simulation studies demonstrate that the ZIBM yields more accurate estimation and reliable inference under conditions commonly observed in microbiome data, compared with existing approaches. An application to a real microbiome study further illustrates its practical utility. These results indicate that the proposed method provides a more flexible and robust statistical framework for mediation analysis involving compositional microbiome data.

Seagull: An Infrastructure for Load Prediction and Optimized Resource Allocation

Microsoft Azure is dedicated to guarantee high quality of service to its customers, in particular, during periods of high customer activity, while controlling cost. We employ a Data Science (DS) driven solution to predict user load and leverage these predictions to optimize resource allocation. To this end, we built the Seagull infrastructure that processes per-server telemetry, validates the data, trains and deploys ML models. The models are used to predict customer load per server (24h into the future), and optimize service operations. Seagull continually re-evaluates accuracy of predictions, fallback to previously known good models and triggers alerts as appropriate. We deployed this infrastructure in production for PostgreSQL and MySQL servers across all Azure regions, and applied it to the problem of scheduling server backups during low-load time. This minimizes interference with user-induced load and improves customer experience.

Minority Voter Distributions and Partisan Gerrymandering

Many people believe that it is disadvantageous for members aligning with a minority party to cluster in cities, as this makes it easier for the majority party to gerrymander district boundaries to diminish the representation of the minority. We examine this effect by exhaustively computing the average representation for every possible $5\times 5$ grid of population placement and district boundaries. We show that, in fact, it is advantageous for the minority to arrange themselves in clusters, as it is positively correlated with representation. We extend this result to more general cases by considering the dual graph of districts, and we also propose and analyze metaheuristic algorithms that allow us to find strong lower bounds for maximum expected representation.
