Exam problems that link physical concepts with electrical phenomena in living cells

We present a collection of biophysics examination problems that focus on electrodiffusion, the superposition principle, and currents in RC circuits, leading towards an understanding of the Hodgkin-Huxley model of action potentials. These problems link basic concepts in electricity to the physiology of living cells. The models described in these problems are intended help students develop an understanding of electrical processes in cells. Our work shows how introductory-level electricity courses can be taught in parallel with basic cell (electro)physiology and how basic concepts of electric circuits apply to cellular phenomena.

Tonic activation of extrasynaptic NMDA receptors decreases intrinsic excitability and promotes bistability in a model of neuronal activity

NMDA receptors (NMDA-R) typically contribute to excitatory synaptic transmission in the central nervous system. While calcium influx through NMDA-R plays a critical role in synaptic plasticity, experimental evidence indicates that NMDAR-mediated calcium influx also modifies neuronal excitability through the activation of calcium-activated potassium channels. This mechanism has not yet been studied theoretically. Our theoretical model provides a simple description of neuronal electrical activity that takes into account the tonic activity of extrasynaptic NMDA receptors and a cytosolic calcium compartment. We show that calcium influx mediated by the tonic activity of NMDA-R can be coupled directly to the activation of calcium-activated potassium channels, resulting in an overall inhibitory effect on neuronal excitability. Furthermore, the presence of tonic NMDA-R activity promotes bistability in electrical activity by dramatically increasing the stimulus interval where both a stable steady state and repetitive firing can coexist. These results could provide an intrinsic mechanism for the constitution of memory traces in neuronal circuits. They also shed light on the way by which $\beta$-amyloids can alter neuronal activity when interfering with NMDA-R in Alzheimer's disease and cerebral ischemia.

The OpenPicoAmp : an open-source planar lipid bilayer amplifier for hands-on learning of neuroscience

Neuroscience education can be promoted by the availability of low cost and engaging teaching materials. To address this issue, we developed an open-source lipid bilayer amplifier, the OpenPicoAmp, which is appropriate for use in introductory courses in biophysics or neurosciences dealing with the electrical properties of the cell membrane. The amplifier is designed using the common lithographic printed circuit board fabrication process and off-the-shelf electronic components. In addition, we propose a specific design for experimental chambers allowing the insertion of a commercially available polytetrafluoroethylene film. This experimental setup can be used in simple experiments in which students monitor the bilayer formation by capacitance measurement and record unitary currents produced by ionic channels like gramicidin A. Used in combination with a low-cost data acquisition board this system provides a complete solution for hands-on lessons, therefore improving the effectiveness in teaching basic neurosciences or biophysics.
