Beyond Weights and Gradients: A Taxonomy of Federated Learning Messages

Federated Learning is rapidly evolving beyond the exchange of traditional model weights and gradients, yet existing definitions fail to capture the full scope of modern payloads like synthetic data and federated analytics. This paper addresses the gap by proposing a formal mathematical definition of a federated message that accounts for both utility and privacy. We introduce a taxonomy that organizes these exchanges into three categories: model structures, statistical summaries, and data-conditioned representations. By evaluating these groups based on computational demands, communication costs, and privacy risks, we provide a clearer understanding of the trade-offs involved in decentralized training. Our review of 202 recent publications highlights a significant shift since 2021 toward diverse messaging paradigms, signaling a move away from standard deep learning updates toward more specialized information sharing. This framework provides a structured path for future research to optimize federated systems for varying hardware and security requirements.

Latent Representation Learning from 3D Brain MRI for Interpretable Prediction in Multiple Sclerosis

We present InfoVAE-Med3D, a latent-representation learning approach for 3D brain MRI that targets interpretable biomarkers of cognitive decline. Standard statistical models and shallow machine learning often lack power, while most deep learning methods behave as black boxes. Our method extends InfoVAE to explicitly maximize mutual information between images and latent variables, producing compact, structured embeddings that retain clinically meaningful content. We evaluate on two cohorts: a large healthy-control dataset (n=6527) with chronological age, and a clinical multiple sclerosis dataset from Charles University in Prague (n=904) with age and Symbol Digit Modalities Test (SDMT) scores. The learned latents support accurate brain-age and SDMT regression, preserve key medical attributes, and form intuitive clusters that aid interpretation. Across reconstruction and downstream prediction tasks, InfoVAE-Med3D consistently outperforms other VAE variants, indicating stronger information capture in the embedding space. By uniting predictive performance with interpretability, InfoVAE-Med3D offers a practical path toward MRI-based biomarkers and more transparent analysis of cognitive deterioration in neurological disease.

Thou Shalt Not Reject the P-value

Since its debut in the 18th century, the P-value has been an important part of hypothesis testing-based scientific discoveries. As the statistical engine accelerates, questions are beginning to be raised, asking to what extent scientific discoveries based on P-values are reliable and reproducible, and the voice calling for adjusting the significance level or banning the P-value has been increasingly heard. Inspired by these questions and discussions, here we enquire into the useful roles and misuses of the P-value in scientific studies. For common misuses and misinterpretations, we provide modest recommendations for practitioners. Additionally, we compare statistical significance with clinical relevance. In parallel, we review the Bayesian alternatives for seeking evidence. Finally, we discuss the promises and risks of using meta-analysis to pool P-values from multiple studies to aggregate evidence. Taken together, the P-value underpins a useful probabilistic decision-making system and provides evidence at a continuous scale. But its interpretation must be contextual, considering the scientific question, experimental design (including the model specification, sample size, and significance level), statistical power, effect size, and reproducibility.
