Instrumental Variable Estimation of Marginal Structural Mean Models for Time-Varying Treatment

Robins 1997 introduced marginal structural models (MSMs), a general class of counterfactual models for the joint effects of time-varying treatment regimes in complex longitudinal studies subject to time-varying confounding. In his work, identification of MSM parameters is established under a sequential randomization assumption (SRA), which rules out unmeasured confounding of treatment assignment over time. We consider sufficient conditions for identification of the parameters of a subclass, Marginal Structural Mean Models (MSMMs), when sequential randomization fails to hold due to unmeasured confounding, using instead a time-varying instrumental variable. Our identification conditions require that no unobserved confounder predicts compliance type for the time-varying treatment. We describe a simple weighted estimator and examine its finite-sample properties in a simulation study. We apply the proposed estimator to examine the effect of delivery hospital on neonatal survival probability.

Consistency of survival tree and forest models: splitting bias and correction

Random survival forest and survival trees are popular models in statistics and machine learning. However, there is a lack of general understanding regarding consistency, splitting rules and influence of the censoring mechanism. In this paper, we investigate the statistical properties of existing methods from several interesting perspectives. First, we show that traditional splitting rules with censored outcomes rely on a biased estimation of the within-node failure distribution. To exactly quantify this bias, we develop a concentration bound of the within-node estimation based on non i.i.d. samples and apply it to the entire forest. Second, we analyze the entanglement between the failure and censoring distributions caused by univariate splits, and show that without correcting the bias at an internal node, survival tree and forest models can still enjoy consistency under suitable conditions. In particular, we demonstrate this property under two cases: a finite-dimensional case where the splitting variables and cutting points are chosen randomly, and a high-dimensional case where the covariates are weakly correlated. Our results can also degenerate into an independent covariate setting, which is commonly used in the random forest literature for high-dimensional sparse models. However, it may not be avoidable that the convergence rate depends on the total number of variables in the failure and censoring distributions. Third, we propose a new splitting rule that compares bias-corrected cumulative hazard functions at each internal node. We show that the rate of consistency of this new model depends only on the number of failure variables, which improves from non-bias-corrected versions. We perform simulation studies to confirm that this can substantially benefit the prediction error.

Marginal Structural Models for Time-varying Endogenous Treatments: A Time-Varying Instrumental Variable Approach

Robins (1998) introduced marginal structural models (MSMs), a general class of counterfactual models for the joint effects of time-varying treatment regimes in complex longitudinal studies subject to time-varying confounding. He established identification of MSM parameters under a sequential randomization assumption (SRA), which essentially rules out unmeasured confounding of treatment assignment over time. In this technical report, we consider sufficient conditions for identification of MSM parameters with the aid of a time-varying instrumental variable, when sequential randomization fails to hold due to unmeasured confounding. Our identification conditions essentially require that no unobserved confounder predicts compliance type for the time-varying treatment, the longitudinal generalization of the identifying condition of Wang and Tchetgen Tchetgen (2018). Under this assumption, We derive a large class of semiparametric estimators that extends standard inverse-probability weighting (IPW), the most popular approach for estimating MSMs under SRA, by incorporating the time-varying IV through a modified set of weights. The set of influence functions for MSM parameters is derived under a semiparametric model with sole restriction on observed data distribution given by the MSM, and is shown to provide a rich class of multiply robust estimators, including a local semiparametric efficient estimator.

Nonparametric generalized fiducial inference for survival functions under censoring

Fiducial Inference, introduced by Fisher in the 1930s, has a long history, which at times aroused passionate disagreements. However, its application has been largely confined to relatively simple parametric problems. In this paper, we present what might be the first time fiducial inference, as generalized by Hannig et al. (2016), is systematically applied to estimation of a nonparametric survival function under right censoring. We find that the resulting fiducial distribution gives rise to surprisingly good statistical procedures applicable to both one sample and two sample problems. In particular, we use the fiducial distribution of a survival function to construct pointwise and curvewise confidence intervals for the survival function, and propose tests based on the curvewise confidence interval. We establish a functional Bernstein-von Mises theorem, and perform thorough simulation studies in scenarios with different levels of censoring. The proposed fiducial based confidence intervals maintain coverage in situations where asymptotic methods often have substantial coverage problems. Furthermore, the average length of the proposed confidence intervals is often shorter than the length of competing methods that maintain coverage. Finally, the proposed fiducial test is more powerful than various types of log-rank tests and sup log-rank tests in some scenarios. We illustrate the proposed fiducial test comparing chemotherapy against chemotherapy combined with radiotherapy using data from the treatment of locally unresectable gastric cancer.

Tree based weighted learning for estimating individualized treatment rules with censored data

Estimating individualized treatment rules is a central task for personalized medicine. [zhao2012estimating] and [zhang2012robust] proposed outcome weighted learning to estimate individualized treatment rules directly through maximizing the expected outcome without modeling the response directly. In this paper, we extend the outcome weighted learning to right censored survival data without requiring either an inverse probability of censoring weighting or a semiparametric modeling of the censoring and failure times as done in [zhao2015doubly]. To accomplish this, we take advantage of the tree based approach proposed in [zhu2012recursively] to nonparametrically impute the survival time in two different ways. The first approach replaces the reward of each individual by the expected survival time, while in the second approach only the censored observations are imputed by their conditional expected failure times. We establish consistency and convergence rates for both estimators. In simulation studies, our estimators demonstrate improved performance compared to existing methods. We also illustrate the proposed method on a phase III clinical trial of non-small cell lung cancer.
