Super-resolution Imaging of the Fluorescent Dipole Assembly with Polarized Structured Illumination Microscopy

Fluorescence polarization microscopy images both the intensity and orientation of fluorescent dipoles, which plays a vital role in studying the molecular structure and dynamics of bio-complex. However, it is difficult to resolve the dipole assemblies on the subcellular structure and their dynamics in living cells with super-resolution. Here we report polarized structured illumination microscopy (pSIM), which decouples the entangled spatial and angular structured illumination through interpreting the dipoles in spatio-angular hyperspace. We demonstrate its application on a series of biological filamentous systems such as cytoskeleton networks and lambda-DNA, and report the dynamics of short actin sliding through myosin-coated surface. Further, pSIM reveals "side-by-side" organization of the actin ring structure in the membrane-associated periodic skeleton in hippocampal neurons. It also images the dipole dynamics of green fluorescent proteins labeled to the microtubules in live U2OS cells. pSIM can be applied directly to a large variety of commercial or home-built SIM systems.

End-to-end Anchored Speech Recognition

Voice-controlled house-hold devices, like Amazon Echo or Google Home, face the problem of performing speech recognition of device-directed speech in the presence of interfering background speech, i.e., background noise and interfering speech from another person or media device in proximity need to be ignored. We propose two end-to-end models to tackle this problem with information extracted from the "anchored segment". The anchored segment refers to the wake-up word part of an audio stream, which contains valuable speaker information that can be used to suppress interfering speech and background noise. The first method is called "Multi-source Attention" where the attention mechanism takes both the speaker information and decoder state into consideration. The second method directly learns a frame-level mask on top of the encoder output. We also explore a multi-task learning setup where we use the ground truth of the mask to guide the learner. Given that audio data with interfering speech is rare in our training data set, we also propose a way to synthesize "noisy" speech from "clean" speech to mitigate the mismatch between training and test data. Our proposed methods show up to 15% relative reduction in WER for Amazon Alexa live data with interfering background speech without significantly degrading on clean speech.

A GPU-based WFST Decoder with Exact Lattice Generation

We describe initial work on an extension of the Kaldi toolkit that supports weighted finite-state transducer (WFST) decoding on Graphics Processing Units (GPUs). We implement token recombination as an atomic GPU operation in order to fully parallelize the Viterbi beam search, and propose a dynamic load balancing strategy for more efficient token passing scheduling among GPU threads. We also redesign the exact lattice generation and lattice pruning algorithms for better utilization of the GPUs. Experiments on the Switchboard corpus show that the proposed method achieves identical 1-best results and lattice quality in recognition and confidence measure tasks, while running 3 to 15 times faster than the single process Kaldi decoder. The above results are reported on different GPU architectures. Additionally we obtain a 46-fold speedup with sequence parallelism and multi-process service (MPS) in GPU.

Detection efficiency evaluation for a large area neutron sensitive microchannel plate detector

In this paper, the detection efficiency of a large area neutron sensitive microchannel plate detector has been evaluated. A 6LiF/ZnS detector was employed as the benchmark detector, the TOF spectra of these two detectors were simultaneously measured and the energy spectra were then deduced to calculate the detection efficiency curve of the nMCP detector. Tests show the detection efficiency@25.3 meV thermal neutron is 34% for this nMCP detector.
