Automated Detection of Myopic Maculopathy in MMAC 2023: Achievements in Classification, Segmentation, and Spherical Equivalent Prediction

Myopic macular degeneration is the most common complication of myopia and the primary cause of vision loss in individuals with pathological myopia. Early detection and prompt treatment are crucial in preventing vision impairment due to myopic maculopathy. This was the focus of the Myopic Maculopathy Analysis Challenge (MMAC), in which we participated. In task 1, classification of myopic maculopathy, we employed the contrastive learning framework, specifically SimCLR, to enhance classification accuracy by effectively capturing enriched features from unlabeled data. This approach not only improved the intrinsic understanding of the data but also elevated the performance of our classification model. For Task 2 (segmentation of myopic maculopathy plus lesions), we have developed independent segmentation models tailored for different lesion segmentation tasks and implemented a test-time augmentation strategy to further enhance the model's performance. As for Task 3 (prediction of spherical equivalent), we have designed a deep regression model based on the data distribution of the dataset and employed an integration strategy to enhance the model's prediction accuracy. The results we obtained are promising and have allowed us to position ourselves in the Top 6 of the classification task, the Top 2 of the segmentation task, and the Top 1 of the prediction task. The code is available at \url{https://github.com/liyihao76/MMAC_LaTIM_Solution}.

Uconnect: Synergistic Spectral CT Reconstruction with U-Nets Connecting the Energy bins

Spectral computed tomography (CT) offers the possibility to reconstruct attenuation images at different energy levels, which can be then used for material decomposition. However, traditional methods reconstruct each energy bin individually and are vulnerable to noise. In this paper, we propose a novel synergistic method for spectral CT reconstruction, namely Uconnect. It utilizes trained convolutional neural networks (CNNs) to connect the energy bins to a latent image so that the full binned data is used synergistically. We experiment on two types of low-dose data: simulated and real patient data. Qualitative and quantitative analysis show that our proposed Uconnect outperforms state-of-art model-based iterative reconstruction (MBIR) techniques as well as CNN-based denoising.

Minimal hypersurfaces for generic metrics in dimension 8

We show that in an $8$-dimensional closed Riemmanian manifold with $C^\infty$-generic metrics, every minimal hypersurface is smooth and nondegenerate. This confirms a full generic regularity conjecture of minimal hypersurfaces in dimension eight. This also enables us to generalize many generic geometric properties of (Almgren-Pitts) min-max minimal hypersurfaces, previously only known in low dimensions, to dimension eight.

Mean Convex Smoothing of Mean Convex Cones

We show that any minimizing hypercone can be perturbed into one side to a properly embedded smooth minimizing hypersurface in the Euclidean space, and every viscosity mean convex cone admits a properly embedded smooth mean convex self-expander asymptotic to it near infinity. These two together confirm a conjecture of Lawson \cite[Problem 5.7]{Brothers86_OpenProblems}.

Generic Regularity of Minimal Hypersurfaces in Dimension 8

In this paper, we show that every $8$-dimensional closed Riemmanian manifold with $C^\infty$-generic metrics admits a smooth minimal hypersurface. This generalized previous results by N. Smale and Chodosh-Liokumovich-Spolaor. Different from their local perturbation techniques, our construction is based on a global perturbation argument in and a novel geometric invariant which counts singular points with suitable weights.

Identification of Repurposable Drugs and Adverse Drug Reactions for Various Courses of COVID-19 Based on Single-Cell RNA Sequencing Data

Coronavirus disease 2019 (COVID-19) has impacted almost every part of human life worldwide, posing a massive threat to human health. There is no specific drug for COVID-19, highlighting the urgent need for the development of effective therapeutics. To identify potentially repurposable drugs, we employed a systematic approach to mine candidates from U.S. FDA-approved drugs and preclinical small-molecule compounds by integrating the gene expression perturbation data for chemicals from the Library of Integrated Network-Based Cellular Signatures project with a publicly available single-cell RNA sequencing dataset from mild and severe COVID-19 patients. We identified 281 FDA-approved drugs that have the potential to be effective against SARS-CoV-2 infection, 16 of which are currently undergoing clinical trials to evaluate their efficacy against COVID-19. We experimentally tested the inhibitory effects of tyrphostin-AG-1478 and brefeldin-a on the replication of the single-stranded ribonucleic acid (ssRNA) virus influenza A virus. In conclusion, we have identified a list of repurposable anti-SARS-CoV-2 drugs using a systems biology approach.

Deformations of Singular Minimal Hypersurfaces I, Isolated Singularities

Locally stable minimal hypersurface could have singularities in dimension $\geq 7$ in general, locally modeled on stable and area-minimizing cones in the Euclidean spaces. In this paper, we present different aspects of how these singularities may affect the local behavior of minimal hypersurfaces. First, given a non-degenerate minimal hypersurface with strictly stable and strictly minimizing tangent cone at each singular point, under any small perturbation of the metric, we show the existence of a nearby minimal hypersurface under new metric. For a generic choice of perturbation direction, we show the entire smoothness of the resulting minimal hypersurface. Second, given a strictly stable minimal hypersurface $Σ$ with strictly minimizing tangent cone at each singular point, we show that $Σ$ is uniquely homologically minimizing in its neighborhood. Lastly, given a family of stable minimal hypersurfaces multiplicity $1$ converges to a stable minimal hypersurface $Σ$ in an eight manifold, we show that there exists a non-trivial Jacobi field on $Σ$ induced by these converging hypersurfaces, which generalizes a result by Ben Sharp in smooth case; Moreover, positivity of this Jacobi field implies smoothness of the converging sequence.

Min-max Minimal Hypersurfaces with Obstacle

We study min-max theory for area functional among hypersurfaces constrained in a smooth manifold with boundary. A Schoen-Simon-type regularity result is proved for integral varifolds which satisfy a variational inequality and restrict to a stable minimal hypersurface in the interior. Based on this, we show that for any admissible family of sweepouts $Π$ in a compact manifold with boundary, there always exists a closed $C^{1,1}$ hypersurface with codimension$\geq 7$ singular set in the interior and having mean curvature pointing outward along boundary realizing the width $\textbf{L}(Π)$.
