A hollow-core fiber based stand-alone multimodal (2-photon, 3-photon, SHG, THG) nonlinear flexible imaging endoscope

Multimodal nonlinear endoscopes have been a topic of intense research over the past two decades, enabling sub-cellular and label-free imaging in areas not reachable with table-top microscopes. They are sophisticated systems that can be implemented on an optical table in a lab environment, but they cannot be easily moved within or out of the lab. We present here a multimodal and flexible nonlinear endoscope system able to perform two photon excited fluorescence and second harmonic generation imaging with a stand-alone and moveable kart integrating a compact ultrashort laser source. In addition, the system can perform three photon excited fluorescence and third harmonic generation thanks to a delivery optical fiber used to deliver ultrashort pulses from massive and not movable laser systems into the stand-alone kart. The endoscopic fiber probes and delivery optical fibers are based on functionalized negative curvature hollow core fibers. The endoscope distal head has a diameter <2.2mm and can perform nonlinear imaging at max 10 frames/s over a field of view up to 600 $\mu$m with a ~1 $\mu$m spatial resolution.

Excitation Enhancement of a Quantum Dot Coupled to a Plasmonic Antenna

Plasmonic antennas are key elements to control the luminescence of quantum emitters. However, the antenna's influence is often hidden by quenching losses. Here, the luminescence of a quantum dot coupled to a gold dimer antenna is investigated. Detailed analysis of the multiply excited states quantifies the antenna's influence on the excitation intensity and the luminescence quantum yield separately.

FCS diffusion laws on two-phases lipid membranes : experimental and Monte-carlo simulation determination of domain size

For more than ten years now, many efforts have been done to identify and characterize nature of obstructed diffusion in model and cellular lipid membranes. Amongst all the techniques developed for this purpose, FCS, by means of determination of FCS diffusion laws, has been shown to be a very efficient approach. In this paper, FCS diffusion laws are used to probe the behavior of a pure lipid and a lipid mixture at temperatures below and above phase transitions, both numerically, using a full thermodynamic model, and experimentally. In both cases FCS diffusion laws exhibit deviation from free diffusion and reveal the existence of domains. The variation of these domains mean size with temperature is in perfect correlation with the enthalpy fluctuation.

A versatile dual spot laser scanning confocal microscopy system for advanced fluorescence correlation spectroscopy analysis in living cell

A fluorescence correlation spectroscopy (FCS) system based on two independent measurement volumes is presented. The optical setup and data acquisition hardware are detailed, as well as a complete protocol to control the location, size and shape of the measurement volumes. A method that allows to monitor independently the excitation and collection efficiency distribution is proposed. Finally, a few examples of measurements that exploit the two spots in static and/or scanning schemes, are reported.

Far-field radiation pattern in Coherent Anti-stokes Raman Scattering (CARS) Microscopy

Far field radiation pattern under tight focusing condition is investigated in Coherent Anti-stokes Raman Scattering (CARS) microscopy both in the forward (F-CARS) and backward (E-CARS) directions. While we assume no refraction index mismatch between the sample and the environing medium, our rigorous numerical electromagnetic computation takes into account the exact polarizations of the excitation laser beams and of the induced nonlinear dipoles. F-CARS and E-CARS radiation patterns, as well as their divergence, are studied as a function of the size of the sample object and compared to the excitation beams.
