Holographic microscopy for the three-dimensional exploration of light scattering from gold nanomarkers in biological media

The 3D structure of light scattering from dark-field illuminated live 3T3 cells marked with 40 nm gold nanomarkers is explored. For this purpose, we use a high resolution holographic microscope combining the off-axis heterodyne geometry and the phase-shifting acquisition of the digital holograms. Images are obtained using a novel 3D reconstruction method providing longitudinally undistorted 3D images. A comparative study of the 3D reconstructions of the scattered fields allows us to locate the gold markers which yield, contrarily to the cellular structures, well defined bright scattering patterns that are not angularly titled and clearly located along the optical axis. This characterization is an unambiguous signature of the presence of the gold biological nanomarkers, and validates the capability of digital holographic microscopy to discriminate them from background signals in live cells.

Imaging Gold Nanoparticles in Living Cells Environments using Heterodyne Digital Holographic Microscopy

This paper describes an imaging microscopic technique based on heterodyne digital holography where subwavelength-sized gold colloids can be imaged in cell environment. Surface cellular receptors of 3T3 mouse fibroblasts are labeled with 40 nm gold nanoparticles, and the biological specimen is imaged in a total internal reflection configuration with holographic microscopy. Due to a higher scattering efficiency of the gold nanoparticles versus that of cellular structures, accurate localization of a gold marker is obtained within a 3D mapping of the entire sample's scattered field, with a lateral precision of 5 nm and 100 nm in the x,y and in the z directions respectively, demonstrating the ability of holographic microscopy to locate nanoparticles in living cells environments.

Metastability Exchange Optical Pumping of Helium-3 at High Pressures and 1.5 T: Comparison of two Optical Pumping Transitions

At low magnetic field, metastability exchange optical pumping of helium-3 is known to provide high nuclear polarizations for pressures around 1 mbar. In a recent paper, we demonstrated that operating at 1.5 T can significantly improve the results of metastability exchange optical pumping at high pressures. Here, we compare the performances of two different optical pumping lines at 1.5 T, and show that either the achieved nuclear polarization or the production rate can be optimized.

High nuclear polarization of helium-3 at low and high pressure by metastability exchange optical pumping at 1.5 Tesla

We perform metastability exchange optical pumping of helium-3 in a strong magnetic field of 1.5 T. The achieved nuclear polarization, from 80% at 1.33 mbar to 25% at 67 mbar, shows a substantial improvement at high pressures with respect to standard low-field optical pumping. The specific mechanisms of metastability exchange optical pumping at high field are investigated, advantages and intrinsic limitations are discussed. From a practical point of view, our results open the way to alternative technological solutions for polarized helium-3 applications and in particular for magnetic resonance imaging of human lungs.
