Federated Learning (FL) faces significant challenges in evolving environments, particularly regarding data heterogeneity and the rigidity of fixed network topologies. To address these issues, this paper proposes \textbf{SOFA-FL} (Self-Organizing Hierarchical Federated Learning with Adaptive Clustered Data Sharing), a novel framework that enables hierarchical federated systems to self-organize and adapt over time. The framework is built upon three core mechanisms: (1) \textbf{Dynamic Multi-branch Agglomerative Clustering (DMAC)}, which constructs an initial efficient hierarchical structure; (2) \textbf{Self-organizing Hierarchical Adaptive Propagation and Evolution (SHAPE)}, which allows the system to dynamically restructure its topology through atomic operations -- grafting, pruning, consolidation, and purification -- to adapt to changes in data distribution; and (3) \textbf{Adaptive Clustered Data Sharing}, which mitigates data heterogeneity by enabling controlled partial data exchange between clients and cluster nodes. By integrating these mechanisms, SOFA-FL effectively captures dynamic relationships among clients and enhances personalization capabilities without relying on predetermined cluster structures.
Chimeric antigen receptor T-cell (CAR-T) therapy represents a paradigm shift in cancer treatment, yet development timelines of 8-12 years and clinical attrition rates exceeding 40-60% highlight critical inefficiencies in target selection, safety assessment, and molecular optimization. We present Bio AI Agent, a multi-agent artificial intelligence system powered by large language models that enables autonomous CAR-T development through collaborative specialized agents. The system comprises six autonomous agents: Target Selection Agent for multi-parametric antigen prioritization across >10,000 cancer-associated targets, Toxicity Prediction Agent for comprehensive safety profiling integrating tissue expression atlases and pharmacovigilance databases, Molecular Design Agent for rational CAR engineering, Patent Intelligence Agent for freedom-to-operate analysis, Clinical Translation Agent for regulatory compliance, and Decision Orchestration Agent for multi-agent coordination. Retrospective validation demonstrated autonomous identification of high-risk targets including FcRH5 (hepatotoxicity) and CD229 (off-tumor toxicity), patent infringement risks for CD38+SLAMF7 combinations, and generation of comprehensive development roadmaps. By enabling parallel processing, specialized reasoning, and autonomous decision-making superior to monolithic AI systems, Bio AI Agent addresses critical gaps in precision oncology development and has potential to accelerate translation of next-generation immunotherapies from discovery to clinic.
Object tracking in inland waterways plays a crucial role in safe and cost-effective applications, including waterborne transportation, sightseeing tours, environmental monitoring and surface rescue. Our Unmanned Surface Vehicle (USV), equipped with a 4D radar, a monocular camera, a GPS, and an IMU, delivers robust tracking capabilities in complex waterborne environments. By leveraging these sensors, our USV collected comprehensive object tracking data, which we present as USVTrack, the first 4D radar-camera tracking dataset tailored for autonomous driving in new generation waterborne transportation systems. Our USVTrack dataset presents rich scenarios, featuring diverse various waterways, varying times of day, and multiple weather and lighting conditions. Moreover, we present a simple but effective radar-camera matching method, termed RCM, which can be plugged into popular two-stage association trackers. Experimental results utilizing RCM demonstrate the effectiveness of the radar-camera matching in improving object tracking accuracy and reliability for autonomous driving in waterborne environments. The USVTrack dataset is public on https://usvtrack.github.io.
Given a compact, oriented, connected surface $F$, we show that the set of connected sutured manifolds $(M,\gamma)$ with $R_{\pm}(\gamma)\cong F$ is generated by the product sutured manifold $(F,\partial F)\times[0,1]$ through surgery triads. This result has applications in Floer theories of $3$--manifolds. The special case when $F=D^2$ or $S^2$ has been a folklore theorem, which has already been used by experts before.

Autonomous driving on water surfaces plays an essential role in executing hazardous and time-consuming missions, such as maritime surveillance, survivors rescue, environmental monitoring, hydrography mapping and waste cleaning. This work presents WaterScenes, the first multi-task 4D radar-camera fusion dataset for autonomous driving on water surfaces. Equipped with a 4D radar and a monocular camera, our Unmanned Surface Vehicle (USV) proffers all-weather solutions for discerning object-related information, including color, shape, texture, range, velocity, azimuth, and elevation. Focusing on typical static and dynamic objects on water surfaces, we label the camera images and radar point clouds at pixel-level and point-level, respectively. In addition to basic perception tasks, such as object detection, instance segmentation and semantic segmentation, we also provide annotations for free-space segmentation and waterline segmentation. Leveraging the multi-task and multi-modal data, we conduct benchmark experiments on the uni-modality of radar and camera, as well as the fused modalities. Experimental results demonstrate that 4D radar-camera fusion can considerably improve the accuracy and robustness of perception on water surfaces, especially in adverse lighting and weather conditions. WaterScenes dataset is public on https://waterscenes.github.io.
We prove that the knot Floer complex of a fibered knot detects whether the monodromy of its fibration is right-veering. In particular, this leads to a purely knot Floer-theoretic characterization of tight contact structures, by the work of Honda, Kazez, and Matic. Our proof makes use of the relationship between the Heegaard Floer homology of mapping tori and the symplectic Floer homology of area-preserving surface diffeomorphisms. We describe applications of this work to Dehn surgeries and taut foliations.
Let $K\subset S^3$ be a hyperbolic fibered knot such that $S^3_{p/q}(K)$, the $\frac pq$--surgery on $K$, is non-hyperbolic. We prove that if the monodromy of $K$ is right-veering, then $0\le\frac pq\le 4g(K)$. The upper bound $4g(K)$ cannot be attained if $S^3_{p/q}(K)$ is a small Seifert fibered L-space. If the monodromy of $K$ is neither right-veering nor left-veering, then $|q|\le3$. As a corollary, for any given positive torus knot $T$, if $p/q\ge4g(T)+4$, then $p/q$ is a characterizing slope. This improves earlier bounds of Ni--Zhang and McCoy. We also prove that some finite/cyclic slopes are characterizing. More precisely, $14$ is characterizing for $T_{4,3}$, $17$ is characterizing for $T_{5,3}$, and $4n+1$ is characterizing for $T_{2n+1,2}$ except when $n=5$. By a recent theorem of Tange, this shows that $T_{2n+1,2}$ is the only knot in $S^3$ admitting a lens space surgery while the Alexander polynomial has the form $t^n-t^{n-1}+t^{n-2}+\text{lower order terms}$. In the appendix, we prove that if the rank of the second term of the knot Floer homology of a fibered knot is $1$, then the monodromy is either right-veering or left-veering.
If $K$ is a fibered knot in a closed, oriented $3$--manifold $Y$ with fiber $F$, and $\widehat{HFK}(Y,K,[F], g(F)-1;\mathbb Z/2\mathbb Z)$ has rank $r$, then the monodromy of $K$ is freely isotopic to a diffeomorphism with at most $r-1$ fixed points. This generalizes earlier work of Baldwin--Hu--Sivek and Ni. We also clarify a misleading formula in Cotton-Clay's computation of the symplectic Floer homology of mapping classes of surfaces.
We prove that if $K$ is a nontrivial null-homotopic knot in a closed oriented $3$--manfiold $Y$ such that $Y-K$ does not have an $S^1\times S^2$ summand, then the zero surgery on $K$ does not have an $S^1\times S^2$ summand. This generalizes a result of Hom and Lidman, who proved the case when $Y$ is an irreducible rational homology sphere.
Using an argument of Baldwin--Hu--Sivek, we prove that if $K$ is a hyperbolic fibered knot with fiber $F$ in a closed, oriented $3$--manifold $Y$, and $\widehat{HFK}(Y,K,[F], g(F)-1)$ has rank $1$, then the monodromy of $K$ is freely isotopic to a pseudo-Anosov map with no fixed points. In particular, this shows that the monodromy of a hyperbolic L-space knot is freely isotopic to a map with no fixed points.
A slope $\frac pq$ is called a characterizing slope for a given knot $K_0\subset S^3$ if whenever the $\frac pq$--surgery on a knot $K\subset S^3$ is homeomorphic to the $\frac pq$--surgery on $K_0$ via an orientation preserving homeomorphism, then $K=K_0$. In a previous paper, we showed that, outside a certain finite set of slopes, only the negative integers could possibly be non-characterizing slopes for the torus knot $T_{5,2}$. Applying recent work of Baldwin--Hu--Sivek, we improve our result by showing that a nontrivial slope $\frac pq$ is a characterizing slope for $T_{5,2}$ if $\frac pq>-1$ and $\frac pq\notin \{0,1, \pm\frac12,\pm\frac13\}$. In particular, every nontrivial L-space slope of $T_{5,2}$ is characterizing for $T_{5,2}$. As a consequence, if a nontrivial $\frac pq$-surgery on a non-torus knot in $S^3$ yields a manifold of finite fundamental group, then $|p|>9$.
Let $K$ be a null-homologous knot in a generalized L-space $Z$ with $b_1(Z)\le1$. Let $F$ be a Seifert surface of $K$ with genus $g$. We show that if $\widehat{HFK}(Z,K,[F],g)$ is supported in a single $\mathbb Z/2\mathbb Z$--grading, then \[\mathrm{rank}\widehat{HFK}(Z,K,[F],g-1)\ge\mathrm{rank}\widehat{HFK}(Z,K,[F],g).\]
Let $K$ be a genus $g$ alternating knot with Alexander polynomial $Δ_K(T)=\sum_{i=-g}^ga_iT^i$. We show that if $|a_g|=|a_{g-1}|$, then $K$ is the torus knot $T_{2g+1,\pm2}$. This is a special case of the Fox Trapezoidal Conjecture. The proof uses Ozsváth and Szabó's work on alternating knots.
We say a null-homologous knot $K$ in a $3$--manifold $Y$ has Property G, if the properties about the Thurston norm and fiberedness of the complement of $K$ is preserved under the zero surgery on $K$. In this paper, we will show that, if the smooth $4$--genus of $K\times\{0\}$ (in a certain homology class) in $(Y\times[0,1])\#N\overline{\mathbb CP^2}$, where $Y$ is a rational homology sphere, is smaller than the Seifert genus of $K$, then $K$ has Property G. When the smooth $4$--genus is $0$, $Y$ can be taken to be any closed, oriented $3$--manifold.
Let $G$ be a finite group acting on a connected compact surface $Σ$, and $M$ be an integer homology 3-sphere. We show that if each element of $G$ is extendable over $M$ with respect to a fixed embedding $Σ\rightarrow M$, then $G$ is extendable over some $M'$ which is 1-dominated by $M$. From this result, in the orientable category we classify all periodic automorphisms of closed surfaces that are extendable over the 3-sphere. The corresponding embedded surface of such an automorphism can always be a Heegaard surface.
Every prism manifold can be parametrized by a pair of relatively prime integers $p>1$ and $q$. In our earlier papers, we determined a complete list of prism manifolds $P(p, q)$ that can be realized by positive integral surgeries on knots in $S^3$ when $q<0$ or $q>p$; in the present work, we solve the case when $0<q<p$. This completes the solution of the realization problem for prism manifolds.
