Acousto-optical coherence tomography with a digital holographic detection scheme

Acousto-optical coherence tomography (AOCT) consists in using random phase jumps on ultrasound and light to achieve a millimeter resolution when imaging thick scattering media. We combined this technique with heterodyne off-axis digital holography. Two-dimensional images of absorbing objects embedded in scattering phantoms are obtained with a good signal-to-noise ratio. We study the impact of the phase modulation characteristics on the amplitude of the acousto-optic signal and on the contrast and apparent size of the absorbing inclusion.

Shot Noise in Digital Holography

We discuss on noise in heterodyne holography in an off-axis configuration. We show that, for a weak signal, the noise is dominated by the shot noise on the reference beam. This noise corresponds to an equivalent noise on the signal beam of one photoelectron per pixel, for the whole sequence of images used to build the digital hologram.

Digital Holography at Shot Noise Level

By a proper arrangement of a digital holography setup, that combines off-axis geometry with phase-shifting recording conditions, it is possible to reach the theoretical shot noise limit, in real-time experiments. We studied this limit, and we show that it corresponds to 1 photo-electron per pixel within the whole frame sequence that is used to reconstruct the holographic image. We also show that Monte Carlo noise synthesis onto holograms measured at high illumination levels enables accurate representation of the experimental holograms measured at very weak illumination levels. An experimental validation of these results is done.

Exploring shot noise and Laser Doppler imagery with heterodyne holography

Heterodyne Holography is a variant of Digital Holography, where the optical frequencies of signal and reference arms can be freely adjusted by acousto-optic modulators. Heterodyne Holography is an extremely versatile and reliable holographic technique, which is able the reach the shot noise limit in sensitivity at very low levels of signal. Frequency tuning enables Heterodyne Holography to become a Laser Doppler imaging technique that is able to analyze various kinds of motion.

Synthetic-aperture experiment in the visible with on-axis digital heterodyne holography

We have developed a new on-axis digital holographic technique, heterodyne holography. The resolution of this technique is limited mainly by the amount of data recorded on two-dimensional photodetectors, i.e., the number of pixels and their size. We demonstrate that it is possible to increase the resolution linearly with the amount of recorded data by aperture synthesis as done in the radar technique but with an optical holographic field.

3D exploration of light scattering from live cells in the presence of gold nanomarkers using holographic microscopy

In this paper, we explore the 3D structure of light scattering from dark-field illuminated live 3T3 cells in the presence of 40 nm gold nanomarkers. For this purpose, we use a high resolution holographic microscope combining the off-axis heterodyne geometry and the phase-shifting acquisition of the digital holograms. A comparative study of the 3D reconstructions of the scattered fields allows us to locate the gold markers which yield, contrarily to the cell structures, well defined bright scattering patterns that are not angularly titled and clearly located along the optical axis (z). This characterization is an unambiguous signature of the presence of gold biological nanomarkers, and validates the capability of digital holographic microscopy to discriminate them from background signals in live ce

Numerical heterodyne holography with two-dimensional photodetector arrays

We present an original heterodyne holography method for digital holography that relies on two-dimensional heterodyne detection to record the phase and the amplitude of a f ield. The technique has been tested on objects as much as 13 mm in size. Consistency checks were performed, and high-resolution images were computed. We show the requirement for a spatial f ilter to select properly sampled near-axis photons. Heterodyne holography is superior to off-axis digital holography for both f ield of view and resolution.

Dark-field digital holographic microscopy for 3D-tracking of gold nanoparticles

We present a new technique that combines off-axis Digital Holography and Dark Field Microscopy to track 100nm gold particles diffusing in water. We show that a single hologram is sufficient to localize several particles in a thick sample with a localization accuracy independent of the particle position. From our measurements we reconstruct the trajectories of the particles and derive their 3D diffusion coefficient. Our results pave the way for quantitative studies of the motion of single nanoparticle in complex media.

Theoretical study of Acousto-optical coherence tomography using random phase jumps on US and light

Acousto-Optical Coherence Tomography (AOCT) is variant of Acousto Optic Imaging (called also ultrasonic modulation imaging) that makes possible to get z resolution with acoustic and optic Continuous Wave (CW) beams. We describe here theoretically the AOCT e ect, and we show that the Acousto Optic tagged photons remains coherent if they are generated within a speci c z region of the sample. We quantify the z selectivity for both the tagged photon eld, and for the M. Lesa re et al. photorefractive signal.

Video-rate laser Doppler vibrometry by heterodyne holography

We report a demonstration video-rate heterodyne holography in off-axis configuration. Reconstruction and display of 1 Megapixel holograms is achieved at 24 frames per second, with a graphics processing unit. Our claims are validated with real-time screening of steady-state vibration amplitudes in a wide-field, non-contact vibrometry experiment.

Holographic laser Doppler ophthalmoscopy

We report laser Doppler ophthalmoscopic fundus imaging in the rat eye with near-IR heterodyne holography. Sequential sampling of the beat of the reflected radiation against a frequency-shifted optical local oscillator is made onto an array detector. Wide-field maps of fluctuation spectra in the 10 Hz to 25 kHz band exhibit angiographic contrasts in the retinal vascular tree without requirement of an exogenous marker.

Fringe free holographic measurements of large amplitude vibrations

In the measurement of the amplitude of vibration of objects, holographic imaging techniques usually involve fringe counting; due to the limited resolution of images, measurements of large amplitudes are not accessible. We demonstrate a technique that suppresses the necessity of fringe counting: frequency sideband imaging, where the order of the sideband is considered as a marker of the amplitude. The measurement is completely local: no comparison with another reference point on the object is necessary. It involves a sharp variation of a signal, which makes it robust against perturbations. The method is demonstrated in an experiment made with a vibrating clarinet reed; phase modulations as large as 1000 radians have been measured.

Parallel heterodyne detection of dynamic light scattering spectra from gold nanoparticles diffusing in viscous fluids

We developed a microscope intended to probe, using a parallel heterodyne receiver, the fluctuation spectrum of light quasi-elastically scattered by gold nanoparticles diffusing in viscous fluids. The cutoff frequencies of the recorded spectra scale up linearly with those expected from single scattering formalism in a wide range of dynamic viscosities (1 to 15 times water viscosity at room temperature). Our scheme enables ensemble-averaged optical fluctuations measurements over multispeckle recordings in low light, at temporal frequencies up to 10 kHz, with a 12 Hz framerate array detector.

Imaging Gold Nanoparticles in Living Cells Environments using Heterodyne Digital Holographic Microscopy

This paper describes an imaging microscopic technique based on heterodyne digital holography where subwavelength-sized gold colloids can be imaged in cell environment. Surface cellular receptors of 3T3 mouse fibroblasts are labeled with 40 nm gold nanoparticles, and the biological specimen is imaged in a total internal reflection configuration with holographic microscopy. Due to a higher scattering efficiency of the gold nanoparticles versus that of cellular structures, accurate localization of a gold marker is obtained within a 3D mapping of the entire sample's scattered field, with a lateral precision of 5 nm and 100 nm in the x,y and in the z directions respectively, demonstrating the ability of holographic microscopy to locate nanoparticles in living cells environments.

Spatiotemporal heterodyne detection

We describe a scheme into which a camera is turned into an efficient tunable frequency filter of a few Hertz bandwidth in an off-axis, heterodyne optical mixing configuration, enabling to perform parallel, high-resolution coherent spectral imaging. This approach is made possible through the combination of a spatial and temporal modulation of the signal to reject noise contributions. Experimental data obtained with dynamically scattered light by a suspension of particles in brownian motion is interpreted.

Imaging of a vibrating object by Sideband Digital Holography

We obtain quantitative measurements of the oscillation amplitude of vibrating objects by using sideband digital holography. The frequency sidebands on the light scattered by the object, shifted by n times the vibration frequency, are selectively detected by heterodyne holography, and images of the object are calculated for different orders n. Orders up to n=120 have been observed, allowing the measurement of amplitudes of oscillation that are significantly larger than the optical wavelength. Using the positions of the zeros of intensity for each value of n, we reconstruct the shape of vibration the object.

Detection of the tagged or untagged photons in acousto-optic imaging of thick highly scattering media by photorefractive adaptive holography

We propose an original adaptive wavefront holographic setup based on the photorefractive effect (PR), to make real-time measurements of acousto-optic signals in thick scattering media, with a high flux collection at high rates for breast tumor detection. We describe here our present state of art and understanding on the problem of breast imaging with PR detection of the acousto-optic signal.
